Biochemistry - Protein purification


The major purpose of our service group is to support the fellow scientists with knowledge and technical know how in the wide field of biochemistry and purification of a variety of biomolecules, e.g. bacterial fusion proteins, membrane proteins, lipids. We perform and establish purification strategies as well as basic protein biochemical methods (labelling of proteins, peptides and lipids; refolding of denatured proteins; enzymatic analysis).


For the isolation of proteins we take advantage of conventional biochemical methods which include techniques like salt-precipitations, ion-exchange chromatography, hydrophobic interaction chromatography,native isoelectric focusing, native gel electrophoresis, reversed phase chromatography, gelfiltration as well as more sophisticated isolation procedures, e.g. lectin and immuno affinity chromatography.


The main instrumental equipment includes the BioCad SPRINT System (PE Biosystems) which is designed for fast development and optimization of any biomolecular purification, using one working platform for all techniques and samples. Furthermore, our group uses the FPLC-system (Pharmacia) as well as two HPLC instruments (Waters, PrepLC4000 and 626 LC) necessary for preparative and analytical approaches, respectively. For the production of recombinant proteins, bacteria can be grown in a 15 L fermentor (Biostat C, Braun Biotech).
