Talk abstract:
T cell polarization and effector function is largely controlled by antigen-presenting cells (APC) – dendritic cells and macrophages - , which provide key signals including direct antigen presentation and co-stimulation, cytokines, metabolites, and other small messengers and receptor-ligand interactions. In solid tumors, APC functionality is impaired by tumor-derived immunosuppressive signals, resulting in unique phenotypes of tumor-associated APC that differ from their homeostatic counterparts. We have now uncovered regulatory modules expressed by distinct tumor-associated APC; however, how these programs translate into tumor-directed T cell responses, and whether novel myeloid targets can be successfully modulated for cancer immunotherapy still needs to be explored.
